Sparian Biosciences has officially started a Phase I clinical study of SBS-147, a novel aryl epoxamide receptor (AEA) agonist aimed at treating both acute and chronic pain conditions. This trial represents an initial human evaluation of a compound designed to broaden therapeutic options in pain management beyond currently available treatments. SBS-147 has been formulated for oral administration, which aims to enhance dosing flexibility compared with prior generations of similar molecules.

The SBS-147 program is built upon the earlier candidate SBS-1000, which was developed for acute pain management. SBS-1000 has already completed a Phase I single ascending dose (SAD) trial using a parenteral formulation and continues to be developed for acute pain indications. SBS-147 represents the next iteration in chemical design and pharmacological approach, offering an oral route of administration and a broader range of potential applications.
Design and Objectives of the Phase I Trial
The Phase I study of SBS-147 incorporates both single ascending dose (SAD) and multiple ascending dose (MAD) cohorts. The primary goals of the trial are to assess tolerability, pharmacokinetics, and safety in healthy adult participants. The development of SBS-147 from Phase I into Phase II is financially supported by a grant from the National Institutes of Health (NIH) and the National Institute on Drug Abuse (NIDA) under the HEAL Initiative. The awarded funding for this stage totals $15 million USD. This grant constitutes the second phase of public support for this development pathway and represents the fifth NIH/NIDA award received by Sparian Biosciences, bringing the total government funding for its programs to approximately $75 million USD.
Academic Collaboration
Professor Susruta Majumdar of Washington University in St. Louis has contributed to the discovery and development of both SBS-147 and SBS-1000. His participation connects the academic research perspective with the biopharmaceutical development pipeline of the AEA agonist program.
